Main research interests are in understanding gene regulation in bacteria for survival during virulence, nutrient limitation etc. The global regulator 6S RNA is a non-coding regulatory RNA that influences adaptation of the bacteria during stationary phase, sporulation, light adaptation or intracellular bacterial survival. 6S RNA orchestrates the switch in transcription regulation during stress response by modulating the availability of variant RNA polymerase holoenzymes – affecting the stationary phase Sigma factor s38-RNA Polymerase holoenzyme and mainly sequestering the housekeeping Sigma factor s70-RNA polymerase holoenzyme.
We aim to characterise the structures of different 6S RNA- RNA polymerase holoenzyme complexes in different states using single particle Cryo-Electron microscopy as the primary methodology to dissect the molecular mechanisms in functional detail. The structures could aid in future to do structure guided molecular design of new antibacterials based on non-coding RNA.
