Structural Biology and Photosynthesis

We are interested in the structure and function of soluble and integral membrane proteins. Although our studies are driven by curiosity to reveal the molecular mechanisms underpinning healthy organisms and disease processes, they have a substantial impact on industrial biotechnology and health.

Two members of the group (Krauss and Pickersgill) use X-ray crystallography to provide a deep understanding of the molecular basis of protein function. These studies are complemented by NMR analyses (Pickersgill and Viles), single particle transmission electron microscopy (Nield) and synchrotron circular dichroism spectroscopy (Janes).

Recent highlights include the crystal structures of an activated bacterial rhodopsin/Galpha-peptide complex, the first structure of an activated G-protein coupled receptor, and the pilotin from the type II secretion system in complex with a peptide from the outer membrane secretin, the first and only such complex. Viles works to understand the process of misfolding and toxicity in protein misfolding diseases such as Alzheimer’s disease and in particular the role of metal ion in accelerating these processes.

Krauss and Nield work on light harvesting complexes and photosystems, Krauss is perhaps best known for solving the structures of photosystems I and II. They are joined by a number of other scientists who also work on photosynthesis. Ruban and Mullineaux use freeze fracture to study the photosynthetic membrane and Mullineaux fluorescence microscopy to probe the structure and dynamics of membranes and the proteins embedded within.

Ruban uses biophysical techniques to probe light capture while Duffy brings a background in quantum physics to computational studies of photosynthetic processes. Heathcote studies electron flow in photosynthesis.

Milanesi and Roessler are working on the role of membranes in protein function and in EPR spectroscopy, respectively. Main works on protein design and characterization and Bessant is an expert in bioinformatics.
